Nanoparticles (NPs) offer unique properties for biomedical applications, leading to new nanomedicines. Recent examples of advanced nanoparticle-based nanomedicines are COVID-19 RNA vaccines. Regardless of the delivery route of the NPs into the body (intravenous or subcutaneous injection, oral, intranasal, etc.), NPs inevitably come into contact with immune cells, such as macrophages. Macrophages are phagocytizing cells that determine the fate and the lifetime of NPs in relevant biological fluids or tissues, which has consequences for both nanosafety and nanomedicine. The aim of this Special Issue is to cover recent advancements in our understanding of NP-macrophage interactions, with a focus on in vitro models for nanosafety and novel nanomedicine approaches that allow the modulation of the immunological profile of macrophages. The current Special Issue compiles nine papers: seven research articles and two review articles. The original articles include studies on the interaction of different nanomaterials, such as multi-walled carbon nanotubes (MWCNTs), amorphous silica, gold nanoparticles, lipid carriers, and microspheres, with macrophages in different scenarios.

Introduction. Conjugated polymer nanoparticles (CPN) have attracted wide attention for potential applications in bio-imaging and as theranostics due to their unique properties including strong photoluminescence, excellent photostability and broad absorption spectra. Despite the promising applications of CPNs, there are only a few studies in the literature investigating the biocompatibility profile of these systems. In this project the impact of various CPN compositions (core/shell materials) on particle properties, their behaviour in biological environments and their interactions with human blood cells, was investigated. Methods. Poly[(9,9-di-n-octylfluorenyl-2, 7-diyl)-alt-(benzo[2, 1,3]thiadiazol-4,8-diyl)] (F8BT), poly[2,5-di(3 ', 7 '-dimethyloctyl)phenylene-1,4-ethynylene] (PPE) and poly[2- methoxy-5-(2-ethylhexyloxy)-1,4-phenylenevinylene] (MEH-PPV) CPNs were synthesised using mini-emulsion technique utilising sodium dodecyl sulphate (SDS), a charged stabiliser, PEG(0.6kDa)-12-hydroxystearate (Solutol ®HS 15), a steric stabiliser, or a mix of lipidic 1,2- dipalmitoyl-sn-glycero-3-phosphotidylcholine (DPPC) and PEG(2K)phosphotidylethanolamine, as a stabilising shell. Zeta-potential and hydrophilic-interactionchromatography were used to study nanoparticle surface properties and protein corona formation. The impact of surface properties on nanoparticle internalisation, cytokine release and cytotoxicity was investigated in J77 4A.1 cell line, and the interactions of CPN s with human blood components was studied by investigating the affect of the particles on induction of haemolysis, activation and agglomeration of platelets and platelet-monocyte adhesion.
